Plasma pharmacokinetics and muscle disposition of marbofloxacin in chickens

چکیده مقاله:
Background: Reports on the pharmacokinetics of marbofloxacin in birds are scarce, even when it is a useful tool in poultry production. OBJECTIVES: Determining marbofloxacin kinetic parameters in plasma and muscle arrangement after intravenous and oral administrations and establishing its withdrawal period. METHODS: Clinical healthy chickens (1.08±0.22 kg) were used as experimental subjects, formed in groups of 5. In Group A (n= 45) birds received 2 mg/kg of marbofloxacin intravenously while group B (n= 65) was given the same oral dose after fasting for 12 hours. Blood samples in the groups and 5 g of muscle were obtained at different times, in A group up to 24 hours and in B group until 120 hours after application. Once the samples were obtained and treated, marbofloxacin was quantified by HPLC. RESULTS: Marbofloxacin in fasted chickens is almost completely absorbed (F= 97%), mean residence time is moderate (6.9 hours), with a high volume of distribution. Orally, mean residence time is less in plasma (4.9 hours) and the withdrawal time was calculated 2.5 days. CONCLUSIONS: The kinetic results are consistent with those presented by other fluoroquinolones in poultry and are in line with short production cycles in chickens.

Pharmacokinetics, Pharmacodynamics, and Drug Disposition
During the last decade, the application of pharmacokinetic and pharmacodynamic modeling techniques has become an increasingly important aspect of contemporary clinical psychopharmacology (1–5). These techniques have been applied during the process of development of new drug entities as well as for the improved understanding of the clinical actions of drugs that are already marketed. Techniques ...
متن کاملThe roles of cytochrome P450 and P-glycoprotein in the pharmacokinetics of florfenicol in chickens
The effects of three selective oral inhibitors, fluvoxamine (FLU), ketoconazole (KET), and verapamil (VER), on the pharmacokinetics (PK) of florfenicol (FFC) were investigated in chickens. The chickens were administered orally with saline solution (SAL), FLU (60 mg/kg), KET (25 mg/kg), or VER (9 mg/kg) for 7 consecutive days. Florfenicol was given to the chickens at a single dose of 30 mg/kg or...
متن کاملOral Bioavailability and Plasma Disposition of Pefloxacin in Healthy Broiler Chickens
The pharmacokinetics of pefloxacin after single 10 mg/kg BW intravenous (IV) and oral doses were studied in healthy broiler chickens. For 24 h, serial blood samples were obtained after IV and oral administration. Concentrations of pefloxacin and its major metabolite N-demethyl pefloxacin (norfloxacin) were measured by use of high-performance liquid chromatography. The plasma concentrations-time...

The association between physiologically dependent pharmacokinetic parameters (CL(B), T1/2beta, Vd(ss)) of marbofloxacin and body weight was studied in eight animal species based on allometric equation Y = aWb, where 'Y' is the pharmacokinetic parameter, 'W' is body weight, 'a' is allometric coefficient (intercept) and 'b' is the exponent that describes relation between pharmacokinetic parameter...
متن کاملPlasma pharmacokinetics and tissue disposition of novel dextran-methylprednisolone conjugates with peptide linkers in rats.
The plasma and tissue disposition of two novel dextran prodrugs of methylprednisolone (MP) containing one (DMP-1) or five (DMP-5) amino acids as linkers were studied in rats. Single 5-mg/kg doses (MP equivalent) of each prodrug or MP were administered intravenously, and blood and tissue samples were collected. Prodrug and drug concentrations were quantitated using HPLC, and noncompartmental pha...

The pharmacokinetics of levofloxacin following its single intramuscular administration (3 mg/kg) was investigated in six male buffalo calves. Peak plasma level of 2.95 ± 0.13μg/ml was observed at 1 h and the drug level above MIC90 in plasma was detected up to 12 h of administration. The bioavailability was 68.1 ± 5.4% and levofloxacin was bound to the plasma proteins to the extent of 19.1 ± 1....
